The cruise is located before the Daintree Village. Follow the main road north until you see the cruise's signs and then turn right when you spot the driveway.
Please note that Cairns is a one-hour and 30-minute drive, Port Douglas is a 45-minute drive and Mossman is a 30-minute drive.
